Kropk\u0119 nad &#8222;i&#8221; postawi\u0142 Profesor Stefan Ch\u0142opicki (<a href=\"http:\/\/jcet.eu\/new\/o-jcet\/zespol-jcet\/\" target=\"_blank\" rel=\"noopener\">kierownik grupy badawczej farmakologii do\u015bwiadczalnej<\/a> oraz dyrektor JCET), zyskuj\u0105c na koniec roku finansowanie na realizacj\u0119 projektu badawczego w ramach konkursu OPUS 15 organizowanego przez <a href=\"https:\/\/www.ncn.gov.pl\/\" target=\"_blank\" rel=\"noopener\">Narodowe Centrum Nauki (NCN)<\/a> w Krakowie.\u00a0<\/strong><\/p>\n<ul>\n<li>Temat projektu: <em>Nowe mechanizmy dysfunkcji \u015br\u00f3db\u0142onka u myszy E3L.CETP &#8211; unikatowym modelu hiperlipidemii z ludzkim profilem lipoprotein<\/em><\/li>\n<li>Przyznany bud\u017cet: <em>1 887 200 PLN<\/em><\/li>\n<\/ul>\n<p>Witamina K, nale\u017c\u0105ca do witamin rozpuszczalnych w t\u0142uszczach, odgrywa istotn\u0105 rol\u0119 w regulacji proces\u00f3w zakrzepowych oraz w procesach przebudowy ko\u015bci poprzez mechanizmy regulowane przez bia\u0142ka zale\u017cne od witaminy K (VKD). Ostatnie badania epidemiologiczne wskazuj\u0105, \u017ce spo\u017cywanie witaminy K2 &#8211; wywieraj\u0105cej g\u0142\u00f3wnie skutki pozaw\u0105trobowe (ale nie witaminy K1- wywieraj\u0105cej g\u0142\u00f3wnie skutki w\u0105trobowe) &#8211; zmniejsza \u015bmiertelno\u015b\u0107 sercowo-naczyniow\u0105 i ca\u0142kowit\u0105 \u015bmiertelno\u015b\u0107. Tego efektu nie mo\u017cna wyt\u0142umaczy\u0107 znanymi dzisiaj mechanizmami dzia\u0142ania witaminy K2. Nasze wst\u0119pne wyniki wykazuj\u0105 po raz pierwszy na to, \u017ce witamina K2 podawana w ma\u0142ej dawce poprawia czynno\u015b\u0107 \u015br\u00f3db\u0142onka u myszy z mia\u017cd\u017cyc\u0105, mierzon\u0105 <em>in vivo<\/em>. Na podstawie naszych wst\u0119pnych wynik\u00f3w bada\u0144 s\u0105dzimy, \u017ce upo\u015bledzenie endogennej syntezy witaminy K2 w \u015br\u00f3db\u0142onku przyczynia si\u0119 do rozwoju dysfunkcji \u015br\u00f3db\u0142onka w mia\u017cd\u017cycy, a suplementacja witaminy K2 odwraca deficyt endogennej witaminy K2 w \u015br\u00f3db\u0142onku, w konsekwencji poprawia status karboksylacji bia\u0142ek zale\u017cnych od witaminy K i poprawia czynno\u015b\u0107 \u015br\u00f3db\u0142onka. Witamina K2 mo\u017ce odgrywa\u0107 istotn\u0105 rol\u0119 w regulacji czynno\u015bci \u015br\u00f3db\u0142onka, kt\u00f3ra jak dot\u0105d jest nieznana.<\/p>\n<p><a href=\"http:\/\/jcet.eu\/new\/wp-content\/uploads\/2019\/01\/SCh-opus.png\"><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-3412\" src=\"http:\/\/jcet.eu\/new\/wp-content\/uploads\/2019\/01\/SCh-opus.png\" alt=\"\" width=\"810\" height=\"450\" srcset=\"https:\/\/jcet.eu\/new\/wp-content\/uploads\/2019\/01\/SCh-opus.png 810w, https:\/\/jcet.eu\/new\/wp-content\/uploads\/2019\/01\/SCh-opus-300x167.png 300w, https:\/\/jcet.eu\/new\/wp-content\/uploads\/2019\/01\/SCh-opus-768x427.png 768w\" sizes=\"auto, (max-width: 810px) 100vw, 810px\" \/><\/a><\/p>\n<p><!--more-->Zdumiewaj\u0105ce jest, \u017ce pomimo d\u0142ugiej historii bada\u0144, wiedza dotycz\u0105ca roli witaminy K w regulacji czynno\u015bci \u015br\u00f3db\u0142onku jest znikoma, zar\u00f3wno w kontek\u015bcie fizjologii, biochemii, patofizjologii jak i farmakologii \u015br\u00f3db\u0142onka i wielu chor\u00f3b zwi\u0105zanych z dysfunkcj\u0105 \u015br\u00f3db\u0142onka. Uzupe\u0142nienie tej wiedzy w ramach niniejszego projektu, mo\u017ce przynie\u015b\u0107 nie tylko nowe zrozumienie mechanizm\u00f3w dzia\u0142ania egzogennej witaminy K2, oraz regulacji czynno\u015bci \u015br\u00f3db\u0142onka przez endogenn\u0105 witamin\u0119 K2, ale mo\u017ce r\u00f3wnie\u017c otworzy\u0107 nowe perspektywy terapeutyczne wielu chor\u00f3b przebiegaj\u0105cych z dysfunkcj\u0105 \u015br\u00f3db\u0142onka naczyniowego.<\/p>\n<p>W projekcie wykorzystane zostan\u0105 myszy E3L.CETP, unikatowy model hiperlipidemii z ludzkim profilem lipoprotein, w kt\u00f3rym dysfunkcja \u015br\u00f3db\u0142onka rozwija si\u0119 powoli i znacznie wyprzedza rozw\u00f3j mia\u017cd\u017cycy.
